Incorporation of 14-C-labeled acetate into lipid by isolated foam cells and by atherosclerotic arterial intima.

چکیده
The synthesis of lipid by rabbit atherosclerotic intima incubated in vitro has been investigated and compared with that of foam cells isolated from intima] lesions. In the atherosclerotic arterial intima, C-labeled acetate is readily incorporated into the lipid fraction, most of the label being found in the phospholipid and cholesterol ester fatty acid fractions. Minimal incorporation into triglyceride, fatty acid, and cholesterol occurred. The major phospholipid synthesized was lecithin but significant incorporation into phosphatidyl inositol, phosphatidyl ethanolamine, and sphingomyelin also took place. The fatty acids synthesized were predominantly saturated in the phospholipid fraction. More monounsaturated fatty acids were present in the cholesterol ester fatty acids, but in both moieties little polyunsaturated fatty acids were labeled by the acetate. Isolated foam cells were also shown to incorporate C-labeled acetate into both phospholipid and cholesterol ester fatty acids. The foam cells, however, incorporated a significantly higher proportion of acetate into cholesterol ester than did the whole intima. The fatty acids synthesized by the foam cells were more polyunsaturated in both the phospholipid and the cholesterol ester fraction than was the case with the whole intima. The relevance of these findings to the origin of lipid in the atherosclerotic lesion is discussed. ADDITIONAL

متن کاملIncorporation of oleic acid into lipid by foam cells in human atherosclerotic lesions.
The incorporation of C-labeled oleic acid into phospholipid, cholesterol ester, and triglyceride in human arterial intima (normal and atherosclerotic) obtained from kidney transplant donors has been investigated in vitro. Most of the oleic acid was incorporated into phospholipid, in both the normal intima and the atherosclerotic lesion, but a greater proportion of label was diverted to choleste...
متن کاملUptake and Metabolism of "C-Labeled Oleic Acid by Atherosclerotic Lesions
The uptake of C-labeled oleic acid and its incorporation into combined lipids by aortic intimas from normal and cholesterol-fed rabbits has been investigated in vitro. More than five times as much oleic acid was taken up by the atherosclerotic intima as by the normal intima. About twice as much oleic acid was incorporated into phospholipid, and twenty times as much into cholesterol ester by the...
متن کاملSterol synthesis in the human arterial intima.
Intimal sterol synthesis was examined in isolated human arterial segments obtained at surgery or at postmortem examination. The tissues were incubated with acetate-1-(14)C and mevalonate-2-(14)C and the incorporation of these precursors into sterols was determined. Intimal sterols were isolated by multiple chromatographic techniques and purified by bromination and oxidation procedures. The resu...
متن کاملExpression of SR-PSOX, a novel cell-surface scavenger receptor for phosphatidylserine and oxidized LDL in human atherosclerotic lesions.
Receptor-mediated endocytosis of oxidized low density lipoprotein (Ox-LDL) by macrophages and the subsequent foam cell transformation in the arterial intima are key events in early atherogenesis. Recently, we have identified a novel macrophage cell-surface receptor for Ox-LDL by expression cloning from a cDNA library of phorbol 12-myristate 13-acetate-stimulated THP-1 cells, designated as the s...
